Description

Principle of the Assay: This assay employs a two-site sandwich ELISA to quantitative WNT3A in Human serum, plasma. An antibody specific for WNT3A has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any WNT3A present is bound by the immobilized antibody. After removing any unbound substances, a biotin - conjugated antibody specific for WNT3A is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of WNT3A bound in the initial step. The color development is stopped and the intensity of the color is measured.
Background: The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. WNT3A is a member of the WNT gene family. It encodes a protein which shows 96% amino acid identity to mouse Wnt3A protein, and 84% to human WNT3 protein, another WNT gene product. This gene is clustered with WNT14 gene, another family member, in chromosome 1q42 region. WNT3A encodes a deduced 352-amino acid protein containing an N-linked glycosylation site and residues conserved among members of the WNT family. The WNT3A protein shares 84.9% sequence identity with human WNT3
